Good morning! I have two urgent questions! Please reply as soon as possible.
1. In our lab, we usually transfer 4 to 5 embryos. To do the assisted hatching i usually put all the embryos to be transferred in one drop and do the assisted hatching and wait around 30min to 1h for transfer. Is that correct, can i leave the embryo in the same drop (where i made the hatching) or is it necessary to move them to another plate after hatching? or to another drop in the same plate?
2. What is the perfect time needed between hatching and transfer?
I do the hatching using laser, i do around 4 to 5 shots
Thanks in advance, |

"Thank you all for your help.
But I didnt get yet the point why do I need to transfer the embryos back to another fresh drop. I do the assisted hatching using laser in the culture medium: in ISM1 (Medicult) or cleavage (COOK) medium and not in a special medium for hatching."

"Andrea,
You have to transfer embryos to fresh culture media.
Zona thinning is better than drilling You do not damage the embryo that way."

change the media

"Hi,
You need to change media from handling medium (Hatching drop) to culture media and u should keep it back in the incubator if its going to be around half an hour before u transferDr. Dharmesh"

"Hi, Andrea you have not specified how much time you take for hatching? which media you use for hatching? whether you do it with lazer or with acid? whether you do partial zona drilling or complete zona drilling?"

"Hi Mr./Mrs. Andrea;
You can apply after assisted hatching in the same plate but different drop and 30 min. is enough before transfer.
Best regards."
