Brown oocytes

Question:
Hello. A 30 yrs old lady with primary subfertility of 2 1/2 yrs with unexplained infertility underwent IVF with long down regulation protocol and urinary gonadotropins. Oocytes on the day of retrival looked normal.However on day 1 after denudation for fertilization check the oocytes looks brownish.Outof 10 oocytes 4 oocytes showed 2PN there was no cleavage. The images are attached. We would like to know has anybody come across such situation. What could be the cause? What should we do in future, ICSI with pateint eggs or consider ovum donation IVF or AID. Pateint is very upset and wants to go for adoption. Kindly give us your opinion.

"We had same type of case. By IVF, fertilization and cleavage occurred. But all blastomeres were dark brown in colour. ET cancelled. On subsequent cycle by ICSI, we could get best embryos and ET done. She is pregnant carrying twin gestation. Some seminal factors probably the cause for such dark blastomeres. In whatever way sperm is harvested, some seminal factors may persist in final preparation. When you are doing IVF, there are higher chances for such factors to be in contact with oocytes. One important note to make is this particular male patient was under anti alcoholic treatment when he came for the first cycle.

Brownish zygotes

"In unexplained Infertility cases, there shall be some hidden problems or pathology which may creat such problems. In such cases, ICSI is the better option and assisted hatching may also help. The brownish zona shows the patient might have a history of endometriotic cysts in some of her cycles. The endometriosis needs to be ruled out. Try to use recombunant FSH instead of urinary gonadotrophins.
