Clevage arrest

Question:
Hi to everyone. We recently did batch of 7 cases ICSI/IVF. We got Very good 2 pn all grade 1. When they are transfered to clevage media from fertilisation media and observe next day we find all PN disappear but none of them cleaved after 48 hours. What could be the couse of it. Do someone has same experiance. What are the possible measures we can take. Thanks

"1. If you kept the 2 PN zygotes cultured in Cleavage media under oil, which is older and not filtered oil, there is chance of bacterial contamination and block cleavage.
2. Cleavage media has to be opened from fresh bottle or it needs to be filtered for sterility.
One thing is certain that this problem is definitely from the lab. and so your quality controls need be scanned."

"Hi,

if you had really 7 different cases all arrested in PN you have a severe problem in your culture. Beginning from media over temperature over culture condition it could be everything end in stimulation protocoll it could be all.
You really should take a trouble shooting from a person not belonging to the clinic because mosteley you need a distance fronm the things to have an idea what`s real the problem.
