Hello! I have a quick question and need an answer please as soon as possible.
During ICSI, i usually split the eggs into several plates, placing around 6 eggs/ plate. when i finish with the first plate and after transfering the eggs to their apppropriate media, i cant continue working with the same injecting pipette, it gets stuck and stops from aspirating. I usually keep on releasing oil, it becomes better but the aspirations remains slow; so i change it every time i move to the next plate.
What shall i do to keep on working with the same injection pipette? |

"when you remove ICSI plate from the invertoscope, you can replace it withe a dishe containing only culture oil to prevent dehydration of PVP inside the injection pipette.

Suck in media once you finish ICSI and don’t keep long time gap between ICSI. If you are using oil syringe, check any air bubbles in tube before starting ICSI.

"Also your colleague can assist you in loading or reloading of the oocytes from and to the ICSI dishes, that will helps in saving of time. i.e before you finish the injection in the first icsi dish and within a proper time ask your assistant to load a non-injected oocytes into a fresh icsi dish, then give him the the one with you, and he will load them to the culturing media, and so on"

"Hi simply use 2 dishes and so avoid leaving the needle in the air by changing the dishes right away. Postpond transferring the embryos till u finished from them all. You did not mention what the brand of ur injection needle.

icsi pipette flushing

"After you finish with the ICSI, try blowing air with both pipettes still under oil. Don't do this at the drops where the eggs are, try moving your pipettes at the edge of the dish. Once you see bubbles, lift the pipettes out and then suck some air back in. They will be free of PVP and medium, so you can use them again for the same patient. Remember, blow under oil, suck when out of it!"

"Hi,
Once i`ve finished my series of injections and need to replenish / replace a dish, I flush the ICSI pipette with media to remove the PVP.
This should solve the problem :)

"You could try lowering the ICSI pipette into a dish with drops of medium or PVP under oil, to keep the glass moist between dishes. It sounds like the PVP on or in the tip is dehydrating.
