| Hey!! Please can anyone help. Sometimes when i do ICSI, after i enter the egg with the injection pipette and i aspirate some cytoplasm, when i try to release the cytoplasm back, the sperm doesnt get down and stays sticked into the injection pipette. what shall i do to avoid pvp from entering while letting the sperm down in such cases?? |

| I want to buy a mini-incubator for blastocyst culture but I don’t know what to chose since I have two possibilities : one is MINC benchtop incubator from COOC, and the second option is ORGIO/Planer Benchtop Incubator BT37 from MidAtlantic. I have strong recomandation from people how work with Cook and nothing about Orgio but Orgio has a good price. Can somebody give me more details about Orgio in comparation with Cook. | Please can anyone suggest training for an embryologist, for cleavage stage and tropoductum biopsy .... followed by fixatation. Also, what do you suggest for blastocyst biopsy media?? Is it the same as what you are using for Day 3 biopsy?? I tried to do a day 5 biopsy but as soon as i break tropo with needle the Blastocyst collapses !! is this is normal? Finally what should I do with a hatched blastocyst? If it is hatched can I still can do a biopsy? Thank you |

oocyte denudation

| hey!! I am currently having problems in oocyte denudation. I usually rely on chemical (hyaluronidase) and mechanical (moving from large stripper tip diameter to smaller one); but sometimes after moving the oocytes from hyaluronidase in order to complete the mechanical mechanism, they stick to each other and the cumulus become very hard. So they can't be aspirated with a stripper and i rely on syringes tips to release the eggs. How can i use syringes without touching the eggs? is there any specific technique? because sometimes the eggs become dead. |

Stability of pH during media equilibration

| Dear All, I would like to know if anybody has monitored the stability of medium pH during equilibration ( in our laboratory we use G1 v5 PLUS, Vitrolife and pH Meter Version 2 from RI). We have done 4 24-hours measurements: 1) in two cases after 8 hours of equilibration pH-meter showed the minimal value and this reading was stable for next 8 hours. Then pH increased at about 0,03-0,04 in the last 8-hours of measurement. 2) in the next two cases after 8 hours of equilibration pH-meter showed the minimal value but this reading was stable only for 2-3 hours and then the pH increased at about 0,03-0,04 too. Has anybody noticed a similar tendency ? |

artifacts in culture dishes.

| Hi guys. sometimes we see artifacts in our culture dishes. we use BD falcon 7575 to aliquote medium and prepare culture dishes. when you open individually packed 7575 pippettes, lot of particles are produced, which you can see in the image. we suspect we see the same particles in our culture dishes. Anyone having this problem? Pls comment and share your experience on this. |

Air injection system v/s Oil injection system

| Is there any difference in fert. rate with air injection system compared to oil injection system. The difference i have seen is that the aspiration of oolema is much smoother, much better control with oil injection system. The rupture of oolema with air system is very traumatic at times. Really worries. Anybody using Air sytem, have any tips suggestions to share. pls comment on how to ensure convenient and atraumatic injection with air injection system |

| Just read an interesting paper using single continuous media to blastocyst. I noticed that embryos were cultured in a low O2 environment. Does anyone know if low O2 tension helps to minimize release of possible toxic compounds into the media or would the results likely be the same at ambient O2. Any thoughts? |

| I am working in bovine OPU IVP. and I have some problems with fertilization when checking cleavage rate at D2 after IVF where no more than 40% is achieved. I had checked fertilization medium ph at begining and ending (7.6 and 7.5) of 18 hs of IVF (TALP Fertilization medium), semen has been already used before and works well, incubator is pretty new but I dont really know how acurrate is CO2 injection and concentration, bicarbonate concentration in media could be not enough to equilibrate? any suggestions? Thanks in advance |

No Cleavage after Fertilisation.

| Hi, I had this problem recently a couple of times, fert check at 18 hrs post ICSI showed 14 ova had 2pn but at 48 hrs only 7 had reached 4 cell stage, no cleavage in the rest, no pn either. I cultured the embryos further to 72 hrs, then 2 more embryos cleaved to 2 cell stage, no development in the rest. Other patients embryos which were cultured during the same time and in similar conditions progressed very well at 48 hrs. Checked everything from temp to Carbon dioxide saturation everything found ok. Second time around same thing occurred, only 50% of fertilised embryos cleaved in one patient whereas other patients embryos developed well. All the patients were given the same drugs and no age factor (both patient below 30 yrs). what may be the reason behind this? |
