Question:
hey!! I am currently having problems in oocyte denudation. I usually rely on chemical (hyaluronidase) and mechanical (moving from large stripper tip diameter to smaller one); but sometimes after moving the oocytes from hyaluronidase in order to complete the mechanical mechanism, they stick to each other and the cumulus become very hard. So they can't be aspirated with a stripper and i rely on syringes tips to release the eggs. How can i use syringes without touching the eggs? is there any specific technique? because sometimes the eggs become dead.

"Hey,
You can always try washing the COCs in gamete buffer before putting them into Hyal.
It actually dilutes n washes away lot of things like excess cumulus cells, follicular fluid etc.
Also you can tease the COC while pick up with the pasteur pipettes leaving behind the oocytes covered with 7-9 layers of cells. It really help in stripping and no effect on fertilization and further development."

"Important points to avoid problems at the time of denudation:
1. collect the egg solely, avoid to transfer follicular fluid with it.
2. collect the eggs in a center well dish in the begining, then try to transfer them to another drop (for example a drop outside the middle circle) and again to another drop, so the eggs will be really free of follicular fluid. You can use the other methods of washing.
2. If it is possible for you, try to cut the extra cumulus with needle.
3. transfer the eggs to a drop of hyalu. and leave them there only for 5 sec. then transfer them to another hyalu. drop and do cleaning by pipetting."
