pH measurement of culture media in clinical IVF

Question:

Dear All,

I wish to gather information on the pH measurement of culture media in clinical IVF laboratories from round the world. I request you all to share your procedure / protocol. I would also appreciate to get responses from those who do not measure pH in their labs.

  1. Do you measure the pH of culture media? How frequently?
  2. What procedure and equipment ( make and type of pH meter and the probe) do you use to measure the pH? Plz brief the procedure.
  3. Do you use different pH at different stages of culture viz. for fertilization, for D1 to D3 culture and for blastocyst culture?
  4. If you use just one brand of media, how frequently do you need to change your incubator settings?

5.a  What is the altitude of your place ?

5.b  Which incubator(s) do you use ? What reading (range of reading may be mentioned) of %CO2 do(es) your incubator(s) display at the desired pH of media ?
