Uneven Blastomeres

Question:
From last 9-10 IVF Cycles im getting an uneven blastomere in the embryos, i have calibrated my CO2 incubator(Hereaus)with 5 percent co2. The media which i have used is Universal IVF media and B2 media and in both the media i have got the uneven blastomeres. Please can u tell me what might be the reason. Thankyou

Re: calibration:

Does you incubator have an infra red sensor (most accurate) or an thermal conductance ( older unit`s & less accurate) ?

Did you calibrate using 5% CO2/95% O2 ? or did you use 5% CO2/5%O2/90% N ? Apparently it makes a difference -i beleive you are meant to use the later.

Did you check your calibration using an independant hand held infra red CO2 device sampling through the sampling port on the front of the inc ? With the conductance type incs i used to find some discrepency and re-calibrate accordingly.

Also, when calibrating the conductance type the inc should have been at a stable 37 C overnight before CO2 calibration was instigated. This is especially true if you are running a humid incubator -as incomplete humidifiction with throw your CO2 calibration out in conductance incubators.

I wish you well. I currently have infra red type incs -but i still check them every morning ( before the doors have been opened) with a hand held IR monitor. Just in case
